Overview

Proline-rich coiled-coil 2C (PRRC2C) is a protein found abundantly in various tissues including adult spinal cord, liver, brain, lung, and ovary. As a key component of stress granules, PRRC2C is linked to cellular responses to stress and appears to regulate gene expression under stress conditions. Its gene is located on chromosome 1q23-24, a region often amplified in cancers. PRRC2C promotes cell proliferation, inhibits apoptosis, and enhances metastasis in multiple tumor types, mainly by upregulating EMT-related proteins like N-cadherin and Vimentin. Knockdown or silencing of PRRC2C reduces tumorigenesis in cellular and mouse models, making it a potential therapeutic and diagnostic target for hepatocellular carcinoma and possibly other cancers. Alternative splicing and mutation of PRRC2C have also been implicated in lung adenocarcinoma and Burkitt lymphoma, respectively, and PRRC2C protein is present in serum from preeclampsia patients, suggesting broader relevance beyond cancer.
