Overview

Proline-rich gamma-carboxyglutamic acid protein 1 (PRRG1) is a vitamin K-dependent, gamma-carboxyglutamic acid (Gla) containing single-pass transmembrane protein. It features a Gla domain at the N-terminus, which requires post-translational gamma-carboxylation by a vitamin K-dependent gamma-carboxylase, and a proline-rich C-terminus containing signaling and cytoskeletal motifs (PPXY and PXXP). PRRG1 is highly expressed in neural tissues, especially the spinal cord, but its precise physiological role is not fully defined. It is not classified as a receptor, enzyme, transporter, or key druggable therapeutic target. Disease associations mainly involve rare hereditary deficiencies of vitamin K-dependent clotting factors, and there are currently no known drugs, biomarkers, or therapeutic safety concerns directly related to PRRG1.
