Proline-rich protein 16 (PRR16), also known as Largen, is a human protein encoded by the PRR16 gene located on chromosome 5q23.1[1][3][5][6]. It consists of 304 amino acids, and its predicted structure is largely disordered except for a short N-terminal α-helix[1]. PRR16 acts as a regulator of cell size, promoting cell size increase independently of canonical mTOR and Hippo signaling pathways; it achieves this by stimulating the translation of specific mRNAs, particularly those affecting mitochondrial functions, thus increasing mitochondrial mass and respiration[6][3][2]. PRR16 is expressed at low levels in various tissues, including skeletal muscle, heart, lung, brain, skin, and bone marrow[1]. It localizes mainly to the nucleus but can also be found in the cytoplasm[1]. There is currently no evidence that PRR16 is a direct therapeutic target, receptor, enzyme, transporter, or transcription factor, and no known drugs directly target this protein[1][3][5][6]. It interacts with several proteins related to cell signaling and protein ubiquitination but has not been conclusively linked to specific diseases as a driver or major biomarker[1][3][5][6].
