Proline-rich protein 33 (PRR33) is a muscle-enriched protein highly expressed in human and mouse skeletal muscle and heart. It acts as a positive regulator of muscle differentiation (myogenesis), is upregulated during myoblast differentiation, and is required for normal myofiber size and skeletal muscle function. PRR33 interacts with DESMIN to coordinate cytoskeleton-mitochondria organization during myogenesis. Genetic removal or knockdown of PRR33 impairs myofiber growth, muscle strength, and muscle regeneration. Although mainly studied for its physiological role in muscle, there is currently no evidence to designate PRR33 as a classical therapeutic target, receptor, enzyme, or transporter[1][2][3][7].
