Overview

Proline-rich protein 5 (PRR5) is a regulatory protein encoded by the PRR5 gene, located on chromosome 22. It comprises a proline-rich domain and serves as an associated subunit of the mTORC2 (mammalian target of rapamycin complex 2), which is a key regulator of cell growth, survival, and cytoskeleton organization in response to growth factor signaling. PRR5 is involved in controlling platelet-derived growth factor receptor beta (PDGFRB) expression and signaling pathways that include Akt and S6K1. Emerging evidence suggests it may act as a tumor suppressor, especially in breast cancer, and is implicated in disease roles ranging from cancer to epilepsy. The function of PRR5 within mTORC2 makes it relevant to therapeutic strategies targeting this signaling axis, although no drugs currently directly target PRR5 in isolation[1][3][6].
