Overview

Proline rich protein BstNI subfamily 2 (PRB2) encodes a member of the basic, proline-rich human salivary glycoprotein family[1][5][7]. The preproprotein is proteolytically processed to produce one or more mature isoforms that are secreted by the parotid glands into saliva[1][2][7]. PRB2 contributes to the highly repetitive, proline-rich composition of saliva, playing roles in oral lubrication, formation of the protective acquired enamel pellicle, and binding of dietary polyphenols, which reduces their astringency and toxicity in the oral cavity[2][7][9]. The gene is polymorphic, with different alleles showing variable numbers of tandem repeats. PRB2 is not considered a classical drug target, receptor, enzyme, transporter, or transcription factor, and no clinically actionable therapeutics are known to interact with it[1][5][7]. Its expression is primarily relevant to oral biology, and while it has been reported in some cancer expression and mutation datasets, it is not established as a disease driver or biomarker[1][5][7].
