Overview

Proline-rich protein HaeIII subfamily 1 (PRH1) is a salivary glycoprotein encoded by the PRH1 gene, predominantly secreted by the parotid and submandibular/sublingual glands. The gene exhibits considerable polymorphism, with several allelic variants (e.g., PIF-s, Pa, Db-s) influencing protein structure and function. PRH1 is a component of the acquired pellicle on teeth, where it acts as a modulator of bacterial adhesion—particularly affecting Streptococcus mutans colonization—and as a potent inhibitor of calcium phosphate crystal growth, helping to protect dental enamel. Certain genetic variants of PRH1 are associated with increased susceptibility to dental caries. PRH1 is not a conventional therapeutic target (i.e., not an enzyme, receptor, or transporter) but is a biomolecule relevant to oral health and disease risk.
