Overview

Prolyl 3-hydroxylase 3 (P3H3) is an enzyme of the leprecan family, classified among the collagen prolyl 3-hydroxylases. It catalyzes the post-translational hydroxylation of specific proline residues in collagen alpha chains, which is important for the proper biosynthesis, folding, and assembly of collagens, particularly type IV and V collagens[1][4][7]. It functions as part of a protein complex in the endoplasmic reticulum and may act redundantly with other prolyl 3-hydroxylases[4][5]. P3H3 gene expression is detected in various tissues and can be downregulated through epigenetic silencing in cancers such as breast carcinoma, where it may act as a tumor suppressor[1][4]. Disease associations include breast cancer and ocular tuberculosis, but direct drug interactions or pharmacological inhibitors/activators are not currently described[4][5]. The enzyme’s disruption may therefore impact extracellular matrix organization, tumor progression, and tissue integrity.
