Overview

Prolyl Hydroxylase Domain-Containing Protein 3 (PHD3) is a cellular oxygen sensor and a member of the prolyl hydroxylase family. It regulates the stability of hypoxia-inducible factors (HIFs) by hydroxylating specific proline residues on HIF-alpha subunits, marking them for proteasomal degradation under normoxic conditions. PHD3 has a broader substrate range than other PHD isoforms, impacting cell motility, apoptosis, and other cellular processes. It acts as a tumor suppressor by promoting apoptosis and inhibiting growth, angiogenesis, differentiation, and metastasis. PHD3 expression is induced under hypoxic conditions and its aggregation behavior is oxygen-dependent.
