Overview

Prolyl Hydroxylase EGLN3 (PHD3) is an enzyme that functions as an oxygen sensor, primarily regulating cellular responses to hypoxia by hydroxylating proline residues in HIF-alpha subunits. This hydroxylation leads to HIF-alpha degradation under normal oxygen conditions and stabilization under hypoxic conditions. It is also involved in apoptosis regulation and DNA damage response. EGLN3 has clinical relevance as a biomarker and potential therapeutic target in various cancers.
