Overview

Prominin-2 is a pentaspan membrane glycoprotein and the second member of the prominin family, structurally related to Prominin-1 (PROM1)[1][2]. It is predominantly expressed in basal epithelial cells and implicated in the organization of plasma membrane microdomains[2][3]. Prominin-2 is involved in the formation of plasma membrane protrusions, cholesterol binding, and recruitment, as well as in the regulation of membrane architecture, possibly through effects on caveolin-1 phosphorylation and lipid rafts[1][3]. It is primarily found in epithelial tissues, including distal tubules and collecting ducts of the kidney[6]. PROM2 is overexpressed in several cancers (including breast, brain, lung, renal, ovarian, tongue cancers, and melanoma) and may contribute to cancer progression and metastatic potential[1][3]. The functions of Prominin-2, and its regulation—by proteins such as androgen receptor—are still being investigated, but PROM2 and PROM1 share similar patterns of gene correlation and may participate in related signaling pathways[1]. No interacting drugs, direct mechanisms of action, or distinct safety concerns have yet been documented specifically for Prominin-2 as a direct therapeutic target in the available sources.
