The **promoter-associated ncRNA regulator of CCND1 (PNCRNA-D)**, also known as **pncRNA-D**, is a 602-nucleotide long noncoding RNA (lncRNA) transcribed from the promoter region of the human cyclin D1 (CCND1) gene[1][2][3][7][9]. It does not encode a protein but functions by interacting with the RNA-binding protein TLS/FUS. This interaction leads to TLS/FUS inhibiting the histone acetyltransferase activity of CBP/p300 at the CCND1 promoter, thereby repressing CCND1 transcription[1][7]. PNCRNA-D expression is upregulated by cellular stresses like irradiation or osmotic shock, and its regulatory activity is modulated through N6-methyladenosine (m^6^A) RNA modification. Reduced m^6^A methylation of PNCRNA-D under stress conditions increases its interaction with TLS/FUS, leading to greater suppression of cyclin D1 and cell cycle arrest at G0/G1 phase[1][2]. PNCRNA-D is a key regulator of cell proliferation and likely plays a role in tumor suppression; it is not currently a recognized therapeutic target or associated with any drugs or clinical biomarkers[1][2][7].
