Overview

PARTICL (Promoter of MAT2A antisense radiation-induced circulating long non-coding RNA) is a human long non-coding RNA (lncRNA) transcribed in the antisense direction within the promoter region of the Methionine adenosyltransferase 2A (MAT2A) gene. It acts as an epigenetic mediator, recruiting Polycomb Repressive Complex 2 (PRC2, notably SUZ12 and EZH2 subunits) and DNA methyltransferase 1 (DNMT1) to form triplex RNA-DNA structures at CpG islands, particularly within MAT2A and WWOX tumor suppressor gene promoters. PARTICL's expression is highly responsive to ionizing radiation, which can transiently increase its levels, leading to enhanced H3K27 trimethylation and DNA methylation at specific genomic loci. This activity results in the silencing of tumor suppressor genes and modifications in the cellular response to DNA damage. Although PARTICL is associated with cancer-related epigenetic regulation, there is currently no evidence it is directly targeted by drugs, nor is it classified as a conventional pharmacological target. High PARTICL expression is linked to metastasis-free survival in osteosarcoma, suggesting a potential use as a biomarker for prognosis.
