Promoting weight loss is a clinical objective and physiological outcome aimed at reducing body mass and adiposity, primarily to treat obesity and its associated metabolic complications [1, 8]. It is not a single therapeutic target (such as a receptor or enzyme) but a complex biological result achieved by modulating multiple pathways involved in energy balance, satiety, and nutrient absorption [2, 10]. Pharmacological interventions to promote weight loss typically involve targeting specific molecules like the Glucagon-like peptide 1 receptor (GLP-1R) to induce satiety and slow gastric emptying, or inhibiting gastric and pancreatic lipases to reduce fat absorption [1, 3, 6]. Effective weight management is a cornerstone for reducing the risk of chronic conditions, including type 2 diabetes mellitus, cardiovascular disease, and nonalcoholic fatty liver disease [5, 10]. Recent therapeutic breakthroughs include multi-receptor agonists, such as tirzepatide (targeting both GLP-1R and GIPR), which have demonstrated the ability to produce substantial and sustained weight loss in clinical populations [3, 8, 10].
Weight loss is promoted through diverse pharmacological mechanisms: GLP-1 and GIP receptor agonists increase satiety and reduce appetite via central nervous system pathways [1, 3, 6]; lipase inhibitors prevent the hydrolysis and subsequent absorption of dietary fats in the gut [8]; and certain agents stimulate energy expenditure by activating thermogenic pathways in brown or beige adipose tissue [4, 5].
