PROP paired-like homeobox 1 (PROP1) is a pituitary-specific paired-like homeodomain transcription factor found exclusively in the pituitary gland, where it is essential for the development, differentiation, and function of hormone-producing cell types in the anterior and intermediate pituitary[1][2][5][7]. PROP1 acts early in pituitary organogenesis, initiating the specification and differentiation process of hormone-secreting cells such as somatotropes, lactotropes, thyrotropes, and gonadotropes by controlling the expression of critical downstream regulators like POU1F1 (Pit-1)[2][5][6]. Mutations in PROP1 are the major genetic cause of combined pituitary hormone deficiency, resulting in deficiencies of growth hormone, thyroid-stimulating hormone, prolactin, gonadotropins, and sometimes adrenal insufficiency[1][2][5][6][7]. PROP1 is not a therapeutic drug target but is clinically significant for genetic diagnosis of specific forms of hypopituitarism.
