Overview

The prostacyclin receptor (IP), encoded by the PTGIR gene, is a G protein-coupled receptor that binds prostacyclin (prostaglandin I2 or PGI2). Activation of the IP receptor leads to vasodilation, inhibition of platelet aggregation, and moderation of vascular smooth muscle cell function, contributing to cardioprotection. It is a therapeutic target for pulmonary arterial hypertension and other diseases involving vasoconstriction or thrombosis. Dysfunction or reduced signaling can increase cardiovascular risk.

Mechanism of action

Agonists bind to the IP receptor, activating Gs proteins, stimulating adenylate cyclase, increasing intracellular cAMP levels, and leading to downstream effects mediated by PKA and Epac.
