Overview

The Prostaglandin E2 (PGE2) biosynthesis pathway is a series of enzymatic reactions that convert arachidonic acid into PGE2, a key signaling molecule involved in inflammation, immune regulation, and various other physiological processes. The pathway involves the action of phospholipase A2 to release arachidonic acid, followed by cyclooxygenase (COX) enzymes (COX-1 and COX-2) to produce prostaglandin H2 (PGH2), and finally prostaglandin E synthases (PGES) to convert PGH2 to PGE2. PGE2 exerts its effects by binding to EP1-EP4 receptors.

Mechanism of action

NSAIDs inhibit COX-1 and COX-2 enzymes, reducing the production of prostaglandins including PGE2; COX-2 selective inhibitors specifically block COX-2; Prostaglandin analogs mimic the effects of PGE2 or other prostaglandins.
