The prostaglandin synthesis pathway refers to the series of biochemical reactions that convert membrane phospholipids into prostaglandins, a group of lipid compounds with diverse physiological roles including inflammation, pain signaling, and regulation of blood flow. This pathway is present in nearly all nucleated cells and is tightly regulated due to the potent biological effects of its products. Key enzymes include Phospholipase A2 (PLA2), Cyclooxygenases (COX-1/2), and various terminal synthases. Products include prostaglandins (PGD2, PGE2, PGF2α), prostacyclin (PGI2), and thromboxane A2 (TXA2). The pathway is targeted by NSAIDs and steroidal anti-inflammatories.
