Prostanoid DP1 receptor (DP1)

Target
DP1
Molecular classification
G protein-coupled receptor (GPCR), Class A GPCR, Rhodopsin-like receptor, Prostanoid receptor

Overview

The Prostanoid DP1 receptor (DP1), also known as PTGDR1, is a G protein-coupled receptor that binds prostaglandin D2 (PGD2). It is expressed in various tissues, including immune cells, cells involved in allergic/inflammatory responses, and other tissues like the gastrointestinal tract. DP1 couples primarily to Gs proteins, leading to increased cAMP levels. It plays complex roles in immune regulation, allergic disorders, pruritus, and reproduction, making it a potential therapeutic target for various diseases, but with significant safety considerations due to its context-dependent effects.

Mechanism of action

Activation of DP1 by agonists leads to Gs protein activation, increased adenylate cyclase activity, and increased intracellular cAMP levels.

Safety considerations

Net effect of targeting DP1 depends strongly on disease context.Activation can be protective or deleterious depending on timing/cell type involved.Excessive activation may exacerbate inflammatory cell recruitment under certain conditions.
