Overview

Prostate and breast cancer overexpressed 1 protein (PBOV1) is a human-specific protein of unknown but cancer-related function[2][4]. The gene is mapped to chromosome 6q23-24 and encodes a 135-amino acid protein from a single-exon transcript[2][3]. PBOV1 is notably upregulated in various cancers—prostate, breast, ovarian, bladder, liver, and glioma—while having minimal to no expression in normal tissues[2][4][3][5]. PBOV1 promotes cancer cell proliferation and tumorigenicity in vitro by enhancing cell cycle progression from G1 to S phase, suppressing cell cycle inhibitors (P21, P27), and upregulating cyclin D1 and retinoblastoma (Rb) protein phosphorylation[1][3]. Its tumor-specific expression profile and regulation by oncogenic pathways (e.g., Hedgehog signaling) make it a promising candidate for biomarker development and, potentially, as a future therapeutic target. However, no clinically approved drugs or therapies directly target PBOV1 to date[4][3][1]. The exact molecular function remains under investigation, and conflicting evidence exists regarding its role as an oncogene or tumor suppressor in different cancers[5][3].
