Prostate-associated gene 3 protein (PAGE3)

Target
PAGE3
Molecular classification
Other (Cancer/testis antigen; non-enzymatic, non-receptor protein)
01

Overview

Prostate-associated gene 3 protein (PAGE3) is a member of the cancer/testis antigen family, typically expressed in a range of tumors and normally limited in expression to certain reproductive tissues. PAGE3 is not an enzyme, receptor, or transporter, but rather a non-enzymatic antigen found in various tumors, where it may play roles in cellular stress responses and the regulation of cell proliferation. Its restricted normal tissue expression and upregulation in cancer make it a potential biomarker and immunotherapeutic target for cancers, especially prostate cancer[3]. No small-molecule drugs are known to directly target PAGE3, but it is being investigated as a cancer biomarker and possible target for T-cell-based immunotherapy.

Mechanism of action

Not established (target is not a common enzyme or receptor; function is antigenic and regulatory)

03

Biological functions

Tumor antigenCellular stress responseRegulation of cell proliferation
04

Disease associations

Cancer (notably prostate cancer and other tumors)
05

Safety considerations

Limited safety data; as a cancer/testis antigen, off-target effects in immunotherapies are a plausible concern if expressed in non-tumor tissues
06

Biomarkers

Tissue expression of PAGE3 is considered a biomarker for some tumors, including prostate cancer
