Overview

Prostate cancer-associated long non-coding RNA on chromosome 7 (PCAL7), also known as FAM131B antisense RNA 2 (FAM131B-AS2), is a long non-coding RNA highly upregulated in prostate cancer and closely correlated with androgen receptor (AR) signaling. PCAL7 is primarily located in the cytoplasm of AR-dependent prostate cancer cells and promotes both cell proliferation and migration. Mechanistically, PCAL7 interacts with and stabilizes Huntingtin-interacting protein 1 (HIP1), which in turn sustains AR signaling by protecting HIP1 from proteasomal degradation. Knockdown of PCAL7 with antisense oligonucleotides not only impairs AR pathway activity but also suppresses tumor growth in xenograft models, highlighting PCAL7 as a potential therapeutic target and biomarker for advanced prostate cancer[1][2]. There are currently no known small-molecule drugs directly targeting PCAL7, but nucleic acid-based therapeutics such as antisense oligonucleotides have demonstrated preclinical efficacy in targeting this lncRNA[2].
