Prostate-specific transcript 1 (non-protein coding), abbreviated as PCGEM1, is a long non-coding RNA (lncRNA) located on chromosome 2q32, predominantly and highly specifically expressed in prostate tissue[1][3]. PCGEM1 is overexpressed in a significant proportion of prostate cancers, particularly in androgen receptor-positive and aggressive tumors[1][2][5]. Functionally, PCGEM1 acts as a regulatory RNA, enhancing cell proliferation, inhibiting apoptosis, and reprogramming metabolism by acting as a coactivator for both the androgen receptor and c-Myc—a major transcription factor in cancer metabolism[2]. PCGEM1 regulates transcriptional networks involved in glucose and nucleotide metabolism[2] and downregulates tumor-suppressive microRNAs such as miR-148a[5]. Despite high prostate specificity and tumor association, its precise prognostic or therapeutic utility remains under investigation, and no drugs directly target PCGEM1 to date[4].
