Overview

Prostate tumor-associated antigens (PTAAs) represent a diverse group of proteins that are overexpressed in prostate malignancy, serving as critical focal points for diagnostic imaging and therapeutic intervention. The most prominent members of this class include Prostate-specific membrane antigen (PSMA), a type II transmembrane glycoprotein with folate hydrolase activity, and Prostate-specific antigen (PSA), a serine protease used primarily as a serum biomarker. These antigens are utilized in various treatment modalities, such as radioligand therapy (e.g., Pluvicto), which delivers targeted radiation to cancer cells, and cancer vaccines (e.g., Sipuleucel-T), which prime the immune system to recognize prostatic acid phosphatase (PAP). While these targets offer high specificity for prostate tissue, therapeutic challenges include off-target expression in organs like the salivary glands and the potential for tumors to lose antigen expression under selective pressure. Overall, PTAAs are foundational to the modern management of metastatic castration-resistant prostate cancer.
