Overview

Protamine 1 is a 49–51 amino acid arginine-rich nuclear protein expressed in sperm during spermatogenesis and essential for chromatin condensation and DNA packaging[1][4]. It replaces histones in mature sperm, enabling efficient transfer of the paternal genome and epigenetic information to the oocyte upon fertilization. Analysis reveals tissue-specific expression in testis, and aberrant expression of PRM1 is associated with certain cancers as a cancer-testis antigen. PRM1 serves as a critical biomarker for male infertility and is under investigation for its potential roles in cancer diagnostics and therapeutics[1][4][3]. PRM1 is not a receptor, enzyme, transporter, or ion channel, but a chromatin-associated DNA-binding protein functioning as part of the sperm-specific nuclear structure[1][4].
