Overview

Protamine 3 is a small, arginine-rich protein expressed in the late stages of spermatogenesis. Its main function is to substitute for histones in sperm chromatin, compacting DNA into a highly condensed and genetically inactive state. This condensation is essential for sperm head morphology and genome stability. Although gene clusters for protamines P1 and P2 are well characterized in mammals, PRM3 (sometimes called 'gene 4') is predicted to have a composition rich in negatively charged amino acids, distinguishing it from other protamines that are positively charged. PRM3 is associated with sperm structure and motility, and mutations may be implicated in male infertility[1][2][3]. No evidence supports PRM3 as a clinically actionable therapeutic target, nor are drugs, biomarkers, or safety issues associated with this molecule currently known.
