Protease-associated domain-containing protein 1 (PRADC1) is a secreted, N-glycosylated protein containing a protease-associated (PA) domain, present across a range of species including humans and mice[1][3][4]. It is widely expressed in metabolically active tissues such as liver, muscle, adipose tissue, heart, kidney, and several brain regions[3][7]. PRADC1 expression is acutely regulated by nutritional state—suppressed by refeeding and altered in obesity—and may play a role in modulating physical activity and fat accumulation, particularly in female mice under high-fat diet challenge[1][3][5]. Although knockout of PRADC1 decreases adiposity and increases physical activity and energy expenditure in mice, it does not improve overall metabolic health under obesogenic conditions[1][3]. The precise physiological and pathological functions of PRADC1 remain incompletely understood, and it is not currently categorized as a therapeutic target or pharmacological biomarker[1][3][4].
