Proteasomal ATPase-associated factor 1 (PAAF1) is a WD repeat-containing protein involved in the regulation of the 26S proteasome, specifically by inhibiting assembly and proteolytic activity through modulation of 19S regulatory complex association with the 20S core particle[1][5]. PAAF1 functions as a negative regulator, ensuring proper assembly dynamics and preventing premature proteasome activity, and plays a unique functional role during HIV-1 infection by promoting proviral transcription through recruitment of the 19S regulatory complex to the viral promoter[1][5]. Alternative splicing produces multiple isoforms; orthologs have been identified in other species. PAAF1's primary molecular function is as a proteasome assembly chaperone, not as a conventional drug target such as a receptor, enzyme, or transporter[1][2][5]. Diseases associated with PAAF1 mutation or dysfunction include Mast syndrome and Xanthinuria type I[1]. No direct gene-specific therapies or drugs are known for PAAF1 at this time.
