Overview

Proteasome 20S subunit beta 6 (PSMB6) is a critical component of the 20S core proteasome, a cylindrical, multi-subunit protease complex present in all eukaryotic cells responsible for ATP- and ubiquitin-dependent degradation of intracellular proteins. PSMB6 is one of seven beta subunits forming the two inner rings of the 20S core, and is associated with caspase-like (peptidyl-glutamyl hydrolase) activity, cleaving after acidic residues. The proteasome is essential for cell homeostasis, cell cycle regulation, antigen processing, and removal of misfolded or damaged proteins. Dysregulation of proteasome activity is implicated in cancer, neurodegenerative diseases, and other disorders, and 20S subunits including PSMB6 are targets for approved anticancer drugs known as proteasome inhibitors.

Mechanism of action

Proteasome inhibitors (e.g., bortezomib, carfilzomib) bind and inhibit the proteolytic activity (caspase-like/caspase-like, chymotrypsin-like, trypsin-like) of the 20S core, blocking substrate protein degradation and leading to apoptosis in rapidly dividing or stressed cells.
