Proteasome assembly chaperone 1 (PSMG1) is an evolutionarily conserved, ubiquitously expressed protein chaperone responsible for promoting the assembly of the α-ring of the 20S core particle (CP) in the eukaryotic proteasome[2][1][3]. PSMG1 functions as part of a heterodimeric complex with PSMG2 (PAC2) to enable proper biogenesis and stability of the proteasome, preventing the formation of non-productive α-ring dimers and ensuring incorporation of subunits during proteasome maturation[3][2][1]. Disruption of PSMG1 leads to proteasome dysfunction and, in animal models, to embryonic lethality, highlighting its fundamental importance in cellular homeostasis[2]. The gene is located on human chromosome 21 and may also be known by aliases relating to Down syndrome due to its chromosomal localization[5][1]. Abnormalities or altered expression of PSMG1 have been linked to processes and diseases including cancer progression, protein quality control disorders, and inflammation[4][1]. Currently, there are no approved drugs targeting PSMG1 directly, and it is not known to interact with small molecule drugs or act as a classical therapeutic target receptor or enzyme[4][1].
