Overview

Proteasome subunit alpha type-6 (PSMA6) is a 27 kDa protein encoded by the PSMA6 gene in humans and functions as one of the essential alpha subunits in the 20S core of the proteasome complex[1][3]. The proteasome is a multi-subunit, ATP-dependent protease responsible for the degradation of most intracellular proteins involved in functions such as protein quality control, cell cycle regulation, apoptosis, and antigen presentation[1][3][4]. PSMA6 forms part of the α-rings that constitute the entrances to the proteolytic chamber and control access of substrate proteins for degradation[1][4]. Dysregulation or mutation in PSMA6 and the proteasome machinery is implicated in cancer, cardiovascular, autoimmune, and neurodegenerative diseases. PSMA6 is targeted indirectly by proteasome inhibitors such as bortezomib, which are used in the treatment of multiple myeloma and other malignancies[3][4].
