Overview

Proteasome subunit beta type-11 (PSMB11/beta5t) is a specialized catalytic subunit incorporated in the thymoproteasome, a variant proteasome found exclusively in cortical thymic epithelial cells. Beta5t uniquely modulates the proteolytic activity of the complex, lowering chymotrypsin-like activity compared to conventional beta5 subunits. This produces a distinct set of self-peptides essential for the positive selection of CD8+ T cells, critical for proper immune system development. In clinical pathology, beta5t serves as a highly specific marker for cortical thymic epithelial cells and is pivotal in distinguishing thymoma subtypes from thymic carcinomas. To date, it is not an established target for approved drugs, but its role as a diagnostic biomarker in thymic neoplasia is well recognized.
