Overview

Proteasome subunit beta type-4 (PSMB4) is a protein encoded by the PSMB4 gene, incorporated into the 20S core of the proteasome complex. As one of 28 subunits, it is critical for the assembly and stability of the proteasome, a multicatalytic proteinase complex responsible for ATP/ubiquitin-dependent degradation of damaged or regulatory proteins in a non-lysosomal pathway. While PSMB4 itself is non-catalytic, its role in forming the proteolytic chamber is essential for the complex's function. Dysregulation of the proteasome system, including PSMB4, is implicated in various pathological states, including cancer, neurodegenerative diseases, cardiac conditions, and inflammation. Therapeutically, the proteasome serves as a validated drug target, particularly in oncology, and circulating proteasome components are emerging as potential disease biomarkers
