Protein aggregation is a molecular phenomenon where proteins, particularly misfolded or intrinsically disordered ones, accumulate and clump together. This can lead to the formation of amorphous aggregates, oligomers, and amyloid fibrils. It is closely linked with numerous diseases, including many neurodegenerative disorders, due to the disruption of cellular processes and proteostasis. Aggregation can be triggered by mutations, environmental stressors, and post-translational modifications, destabilizing native protein conformations and exposing hydrophobic regions.
