Protein-arginine N-methyltransferase 8 (PRMT8) is a member of the protein arginine methyltransferase family, enzymes that catalyze the transfer of methyl groups from S-adenosyl-L-methionine (SAM or AdoMet) to the guanidino nitrogen atoms of arginine residues in proteins. PRMT8 is unique among PRMTs due to its highly restricted expression pattern and membrane association. It plays multifaceted roles in neuronal biology, including regulation of dendritic arborization and growth, control of synapse maturation and plasticity, and involvement in neuronal differentiation. Alterations or loss of function of PRMT8 have been associated with changes in brain phospholipid composition, decreased mitochondrial stress capacity, and increased neuroinflammation after hypoxic injury. Genetic studies suggest involvement in neurodevelopmental disorders and microcephaly.
