Overview

Protein C is a vitamin K-dependent plasma glycoprotein that functions as a major anticoagulant in the blood. It is synthesized in the liver as an inactive zymogen and is activated by the thrombin-thrombomodulin complex on endothelial cells. Activated Protein C (APC) degrades coagulation factors Va and VIIIa, thereby inhibiting further thrombin generation and clot formation. Beyond anticoagulation, APC has anti-inflammatory effects and helps maintain vascular barrier function. Deficiency or dysfunction of Protein C leads to increased risk for abnormal blood clotting disorders.

Mechanism of action

Upon activation by the thrombin-thrombomodulin complex, Protein C becomes activated protein C (APC), which acts as a serine protease anticoagulant by degrading coagulation factors Va and VIIIa, thereby inhibiting further thrombin generation and clot formation.
