Overview

Protein C-mannosyltransferase DPY19L3 (DPY19L3) is a multipass transmembrane enzyme localized to the endoplasmic reticulum and functions as a C-mannosyltransferase, catalyzing the transfer of mannose from dolichylphosphate mannose to tryptophan residues in target proteins that contain the WxxW or WxxC consensus sequence[2][7][1][3]. DPY19L3 specifically mediates C-mannosylation of R-spondin 1 (RSPO1), a regulator of Wnt signaling, and has also been shown to mannosylate the netrin receptor UNC5A, typically at the third tryptophan of WxxWxxWxxC motifs[7][4]. DPY19L3-mediated C-mannosylation governs secretion and biological activity of certain substrates such as RSPO1[1][3]. Functional studies in cancer cell models indicate that DPY19L3 is necessary for vasculogenic mimicry and proliferation of tumor cells, implicating its role in oncogenesis[5]. Mutations in the gene are associated with male infertility due to globozoospermia and spermatogenic failure[7]. There are currently no drugs or specific mechanisms of action targeting DPY19L3 clinically, and there are no established biomarkers or safety concerns directly attributed to modulation of this enzyme. The closely related isoform 2 of DPY19L3 lacks C-mannosyltransferase activity[2][5][7].
