Overview

Protein CROC-4 (also known as C1orf61 or "contingent replication of cDNA 4") is a nuclear protein encoded by the C1orf61 gene, which also serves as the host gene for the primary transcript of miR-9-1, one of the three genes encoding the microRNA miR-9 in humans. The CROC-4 protein acts as a transcriptional activator, particularly of the c-fos proto-oncogene promoter, and is believed to be involved in neuronal development and cellular plasticity, especially during brain formation. Expression of CROC-4/miR-9-1 is highly tissue-specific and is tightly correlated with proliferating and migrating cells in the developing brain. The miR-9-1/CROC-4 locus is induced by inflammatory signals, including lipopolysaccharide (LPS) and TNF-α, via NF-κB-dependent activation, serving a key role in the innate immune response by regulating NF-κB signaling through miR-9. miR-9 has additional roles in neuronal function, insulin-producing cell regulation, and cancer pathogenesis. However, Protein CROC-4 itself is not a typical drug target or therapeutic receptor, and no specific drugs or direct mechanisms of action involving this protein are currently established. Note: This entry is not a canonical therapeutic target but rather a host gene for a regulatory microRNA. The primary biological relevance for drug discovery relates to the microRNA (miR-9) it encodes and regulates, rather than CROC-4 as a standalone protein.
