Overview

Protein dachsous homolog 1 (DCHS1) is a member of the cadherin superfamily and acts as an atypical cadherin mediating calcium-dependent cell-cell adhesion in multiple tissues[1][2][3][4][6]. Expressed predominantly in fibroblasts, DCHS1 plays critical roles in the regulation of cell polarity, proliferation, and differentiation during development, and is essential for tissue morphogenesis including the heart, kidneys, lungs, brain, and skeleton[2][3][7]. DCHS1 interacts with the receptor FAT4 via heterophilic binding along its extracellular cadherin domains, forming a complex that coordinates planar cell polarity and structural organization in developing tissues[4][2]. Mutations in DCHS1 are clinically significant, causing mitral valve prolapse and syndromic developmental disorders such as Van Maldergem syndrome[3][5]. Its principal molecular function is in mediating cell adhesion and organizing cellular behavior; it is not known to be targeted by approved drugs nor to serve as a drug receptor, but its mutation status can serve as a biomarker for specific syndromic and cardiovascular conditions[3][1].
