Protein disulfide-isomerase TMX3 (TMX3) is an enzyme of the protein disulfide isomerase (PDI) family and part of the thioredoxin superfamily[1][2][3][4]. It is a type I transmembrane protein primarily localized to the endoplasmic reticulum (ER) membrane, encoded by the TMX3 gene on human chromosome 18[1][3][4]. TMX3 contains one redox-active (a-type) thioredoxin-like domain with a CGHC active site motif, followed by two redox-inactive (b- and b'-type) thioredoxin-like domains[1][3]. This multidomain structure contributes to its enzymatic function and structural stability[3]. TMX3 is involved in the formation, reduction, and isomerization of disulfide bonds in nascent membrane proteins, aiding their correct folding and assembly, particularly as a foldase for membrane-associated proteins like certain ligand-gated ion channels (e.g., insect nicotinic acetylcholine receptors)[1]. TMX3 is broadly expressed across human tissues[1]. While there is evidence it may be important in ER quality control and protein maturation, specific disease roles and drug interactions for TMX3 have not been well characterized in the literature to date[1][2][4].
