Overview

Protein EFR3 homolog A is a peripheral membrane protein essential for the localization and function of the PI4KA complex at the plasma membrane, which regulates the synthesis of critical signaling lipids. EFR3A’s roles include controlling cellular responses to GPCRs, organizing RAS nanoclusters for oncogenic signaling, and modulating insulin-stimulated glucose transport. Mutations and dysregulation of EFR3A have been linked to autism spectrum disorders, cancers, and cardiovascular disease, indicating a broad though currently indirect pathophysiological relevance. While pivotal in membrane lipid signaling and cellular homeostasis, EFR3A is not a direct drug target or biomarker in clinical practice.
