Overview

Protein Hook homolog 3 (HOOK3) is a member of the Hook family of cytosolic coiled-coil proteins that possess conserved N-terminal domains mediating attachment to microtubules and divergent C-terminal domains responsible for organelle-specific interactions[2][3][4]. HOOK3 localizes mainly to the Golgi complex, participates in Golgi positioning and organization, and acts as a molecular adaptor linking organelles to the microtubule cytoskeleton[2][3]. It also interacts with cargo motors including dynein/dynactin and KIF1C, activating the latter and scaffolding complexes for bidirectional vesicle transport along microtubules[5]. Although HOOK3 has cellular functions critical for intracellular trafficking and organelle architecture, it is not classified as a canonical therapeutic target such as a receptor, enzyme, transporter, or ion channel[2][4][5]. No interacting drugs, mechanisms of action, recognized roles as a biomarker, or direct disease associations have been established in the literature to date.
