Protein interacting with C kinase 1 (PICK1) is a cytosolic adaptor and scaffold protein distinguished by its PDZ and BAR domains, best known for regulating the trafficking and subcellular localization of diverse membrane proteins, especially the AMPA-type glutamate receptors, at neuronal synapses[1][3][5][7]. PICK1 interacts with protein kinase C alpha (PRKCA), and its activity is critical for synaptic plasticity, learning, and memory, functioning via the clustering and internalization of receptors[1][3]. It has also been implicated in actin cytoskeleton organization and the development of certain forms of male infertility[1]. Recent research has positioned PICK1 as a potential therapeutic target, especially for neuropsychiatric and neurodegenerative disorders, through development of selective inhibitors that disrupt PICK1–GluA2 interactions and associated synaptic dysfunction[5].
