Protein IWS1 homolog (IWS1) is a transcription elongation factor that defines the composition of the RNA polymerase II (RNAPII) elongation complex. It acts as an assembly factor to recruit mRNA export and splicing factors (ALYREF/THOC4, EXOSC10) and the histone modifying enzyme SETD2 to the elongation complex via its binding to SUPT6H, which itself is bound to the CTD of RNAPII. This facilitates efficient mRNA processing and export, as well as elongation-coupled H3K36 trimethylation—a signature chromatin mark of active transcription. IWS1 interactions are essential for proper pre-mRNA splicing, mRNA export, and cell viability. Loss or depletion of IWS1 impairs embryonic development, with major developmental arrest in model systems such as mouse embryos. While it is not a classical receptor or enzyme drug target, it is considered a critical regulatory node in gene expression and chromatin biology.
