Overview

Protein kinase C and casein kinase substrate in neurons protein 1 (PACSIN1), also known as syndapin-1, is a neuronal scaffolding protein that plays a critical role in membrane tubulation, endocytosis, and vesicle-mediated trafficking. By binding to dynamin family GTPases and actin cytoskeleton regulators, PACSIN1 facilitates synaptic vesicle recycling, receptor trafficking (notably AMPA and NMDA receptors), and contributes to synaptic plasticity and morphogenesis[1][2][3][4][5]. Its F-BAR domain mediates membrane curvature and tubulation, while the SH3 domain enables protein-protein interactions essential for endocytosis and actin regulation[2][5]. Although PACSIN1 is connected to several neurological and developmental disorders, it is not classically considered a therapeutic drug target but is widely studied in the context of neuronal physiology and disease[1][2][3].
