Overview

Protein kinase C and casein kinase substrate in neurons protein 3 (PACSIN3, also called syndapin III) is a cytoplasmic protein highly expressed in muscle tissue, where it is necessary for the biogenesis of caveolae—flask-shaped plasma membrane invaginations involved in membrane tension buffering and signaling[2][4][5]. PACSIN3 contains an F-BAR domain, which confers membrane-shaping ability, and an SH3 domain, mediating protein–protein interactions[1][3]. It localizes to the plasma membrane, particularly at transition zones between flat membrane and caveolar invaginations, where it helps structure and stabilize caveolae, especially those marked by caveolin-3 (CAV3) in muscle cells[2][4][5]. Its absence disrupts proper caveolae formation, compromising muscle cell integrity during mechanical stress, and appears linked to certain muscular diseases[2][4][5]. It is not classified as a therapeutic target (e.g., receptor, enzyme, transporter) and no approved drugs directly target it[3][5].
