Overview

Protein kinase C beta isoform (PKC-β) is a member of the protein kinase C family of serine/threonine kinases that regulate diverse cellular functions by phosphorylating target proteins[3][1][2]. PKC-β is a conventional PKC isoform, requiring calcium, diacylglycerol (DAG), and phosphatidylserine for activation[1][3]. It plays a critical role in B cell receptor signaling, immune cell activation, cell proliferation, and survival[5][4]. PKC-β is implicated in various diseases, most notably certain lymphomas, diabetic retinopathy, and cardiovascular disease, and is considered an important therapeutic target in oncology and inflammatory conditions[2][4][8]. Clinical inhibitors such as sotrastaurin, ruboxistaurin, and enzastaurin target PKC-β either directly or as part of broader pan-PKC inhibition, with ongoing research into isoform-specific targeting for improved efficacy and safety[6][8]. PKC-β is monitored as a biomarker in some hematologic malignancies and diabetic complications, but its physiological ubiquity presents challenges for therapeutic selectivity and toxicity[4][6].
