Protein kinase C beta type I (PKC βI) and Protein kinase C delta (PKC δ) are distinct members of the protein kinase C family, a group of serine/threonine kinases involved in critical cellular signaling pathways (UniProt P05771, P09174). PKC βI is a classical, calcium-dependent isoform primarily expressed in B-cells and vascular tissues, where it mediates B-cell receptor signaling and contributes to the microvascular complications of diabetes (PubMed: 15897562). PKC δ is a novel, calcium-independent isoform that plays a complex role in regulating apoptosis, cell cycle progression, and immune responses, often acting as a tumor suppressor or a mediator of inflammation depending on the tissue context (PubMed: 19819944). Therapeutic targeting of these isoforms is explored in oncology and immunology, with drugs like sotrastaurin acting as multi-isoform inhibitors to treat B-cell malignancies and prevent organ transplant rejection (PubMed: 21170018). However, the structural similarity between PKC isoforms makes achieving high selectivity difficult, leading to potential safety concerns such as gastrointestinal distress and broad immunosuppression (PubMed: 23539152).
