Overview

Protein kinase casein kinase II subunit alpha (CK2α) is a ubiquitously expressed, highly conserved serine/threonine protein kinase and the catalytic subunit of the tetrameric CK2 holoenzyme, which is composed of two catalytic (α or α′) and two regulatory (β) subunits[1][3][5]. CK2α phosphorylates hundreds of substrates and regulates key cellular processes including cell cycle progression, signal transduction, apoptosis, and DNA repair[5]. It is constitutively active and plays essential roles in normal physiology and pathology, notably cancer, where it is often overexpressed and associated with poor prognosis[4][6]. CK2α is a validated therapeutic target under active investigation with several small molecule inhibitors in preclinical and clinical development[2][4][5][6]. Its centrality in cellular signaling underlies both its promise and challenges as a drug target, given the potential for widespread biological impact when inhibited.
