Overview

Protein kinase D1 is a stress-activated serine/threonine kinase that functions as an intracellular signaling node, integrating signals from PKC activation and diacylglycerol accumulation. It has a modular structure with N-terminal regulatory domains (including C1A/C1B and PH domains) and a C-terminal kinase domain. PKD1 localizes to the cytosol, nucleus, membranes, Golgi, and mitochondria, executing pivotal roles in signal transduction, regulation of cell proliferation, apoptosis, differentiation, immune modulation, membrane trafficking, and migration. Its activity is essential for normal development, and dysregulation is implicated in cancer progression, metastasis, and other pathologies.
